BLUF: An international research team has made important discoveries about the Candida fungi that, owing to rising global fungal infections, are key to future treatment strategies.

Medical treatments that exacerbate vulnerability to fungal infections are more and more active, leading to a rise in global fungal infections affecting a billion people annually and causing over a million deaths. The treatment of these infections becomes complicated due to emerging resistant strains, the limited variety of existing antifungal drugs, their side effects, and costs. Consequently, there is a pressing need for more effective treatments.

A team led by Dr. Toni Gabaldón, based at the Institute for Research in Biomedicine (IRB Barcelona) and the Barcelona Supercomputing Center, identified many influential genes in six species of the Candida fungus. This ground-breaking research reveals the adaptation of these pathogens to humans and antifungal drugs, bringing valuable insights to develop effective Candida infection treatments.

An analysis of nearly two thousand genomes from six prominent Candida species, obtained from clinical samples, was made. This wide-scale study discovered genetic variants connected to antifungal drug resistance, offering a comprehensive understanding of both known and new resistance mechanisms towards different antifungal drugs. The team’s findings also hint at the potential spread of resistance through mating between susceptible and resistant strains, contributing to the prevalence of drug-resistant Candida pathogens.

The study, contributing significantly to the understanding of these infections, has resulted in a comprehensive catalog of variants and drivers of drug resistance, crucial for future investigations and development of better infection treatments. This work was supported by Spanish and European institutions, and a private foundation.
